What Science Wants to Know

Most scholars agree that Isaac Newton, while formulating the laws of force and gravity and in?venting the calculus in the late 1600s, probably knew all the science there was to know at the time. In the ensuing 350 years an estimated 50 million research papers and innumerable books have been published in the natural sciences and mathematics. The modern high school student probably now possesses more scientific knowledge than Newton did, yet science to many people seems to be an impenetrable mountain of facts.

One way scientists have tried to cope with this mountain is by becoming more and more specialized, with limited success. As a biologist, I wouldn?t expect to get past the first two sentences of a physics paper. Even papers in immunology or cell biology mystify me?and so do some papers in my own field, neurobiology. Every day my expertise seems to get narrower. So scientists have had to fall back on another strategy for coping with the mountain of information: we largely ignore it.

That shouldn?t come as a surprise. Sure, you have to know a lot to be a scientist, but knowing a lot is not what makes a scientist. What makes a scientist is ignorance. This may sound ridiculous, but for scientists the facts are just a starting place. In science, every new discovery raises 10 new questions, as playwright George Bernard Shaw sardonically declared in a dinner toast to Albert Einstein.

By this calculus, ignorance will always grow faster than knowledge. Scientists and laypeople alike ?would agree that for all we have come to know, there is far more we don?t know. More important, everyday there is far more we know we don?t know. One crucial outcome of scientific knowledge is to generate new and better ways of being ignorant: not the kind of ignorance that is associated with a lack of curiosity or education but rather a cultivated, high-quality ignorance. This gets to the essence of what scientists do: they make distinctions between qualities of ignorance. They do it in grant proposals and over beers at meetings. As James Clerk Maxwell, probably the greatest physicist between Newton and Einstein, said, ?Thoroughly conscious ignorance ... is a prelude to every real advance in knowledge.?

This perspective on science?that it is about the questions more than the answers?should come as something of a relief. It makes science less threatening and far more friendly and, in fact, fun. Science becomes a series of elegant puzzles and puzzles within puzzles?and who doesn?t like puzzles? Questions are also more accessible and often more interesting than answers; answers tend to be the end of the process, whereas questions have you in the thick of things. I can?t grasp much of immunology even though I have a fancy Ph.D., but the wonderful thing is that most immunologists can?t either?no one knows everything anymore. I can, however, understand the questions that drive immunology. And although I don?t pretend to understand much about quantum physics, I can appreciate how the questions in that field arise and why they are so fundamental. Emphasizing ignorance is inclusive; it makes everyone feel more equal in the same way the infinity of space pares everyone down to size.

Of late this side of science has taken a backseat in the public mind to what I call the accumulation view of science?that it is a pile of facts way too big for us to ever hope to conquer. But if scientists would talk about the questions rather than boring your eyes out of their sockets with reams of jargon, and if the media reported not only on new discoveries but the questions they answered and the new puzzles they created, and if educators stopped trafficking in facts that are already available on Wikipedia?then we might find a public once again engaged in this great adventure that has been going on for the past 15 generations.

Samsung ships five million Galaxy Notes in just five months

Five is an important number for Samsung's Galaxy Note. It has a 5.3-inch screen and now, just five months since it debuted Samsung has announced it's shipped five million units (the one million mark was crossed right around the end of December). That's certainly long enough for you to decide if you're in love with its super-sized frame or the accompanying S-Pen stylus, but unfortunately it has not been quite enough time for the highly-anticipated Ice Cream Sandwich software update to be released. While the world waits for the Premium Suite of apps and Android 4.0, let's think back to all the good times we had, including its initial unveiling, our original review and finally its arrival in the US on AT&T. So, are you convinced yet that there's a place in the world for a device like this, or five million phablets later are you still thinking this is just a fad?

Updated 4:33 p.m. ET: CHARLESTON, W.Va. --??Eight people, including a woman celebrating her 26th birthday and six young children, were killed early Saturday when fire swept through a two-story home while they slept.

A?seventh child was?also hurt in the blaze, which was reported around 3:30 a.m., and was in critical condition.?

It was not immediately clear what caused the blaze, which?appeared to have started in a front room on the first floor.

When firefighters arrived, the house was fully engulfed in flames, said Bob Sharp, assistant chief for the Charleston Fire Department. They found four people upstairs and five downstairs; all died except for one child. A tenth person escaped to a neighbor's house and called 911, Sharp said.

NBC's Gabe Gutierrez reports.

All of the children who died were 8 years old or younger.

Charleston Mayor Danny Jones said the two-story wood frame home did not have a working smoke detector.

There was one smoke detector in the kitchen but it was installed too low and would have taken too long to activate, Sharp said. Another broken smoke detector was found near the basement - so essentially there were no working detectors.

"That means people couldn't have been alerted," Jones said. "They didn't have a chance."

The victims, some identified by only their first names, were Alisha Carter-Camp, 26; Alex Seal, an adult, age unavailable;?Keahna Camp, 8;?Jeremiah Camp, Elijah Scott, ?3; Kiki, 3; Gigi, 3; and?Emanuel Jones,?18 months.

Carter-Camp lived at the home with her children and her sister's children, WSAZ-V reported.

The sister, Latasha Isabelle-Jones, 24, was outside the home when it started to burn and was not injured. A 7-year-old boy, Bryan Timothy Camp, also survived and was listed in critical condition at a hospital.

"We have reason to believe that a lot of the people stayed in the house more than one night and maybe on a weekly basis," Jones said. "These people may have had residences in other places, but a lot of people lived in this house."

Carter-Camp's 26th birthday was Saturday, and a party was being held at the home for her, authorities said.

People started showing up for the party around 2 p.m. Friday and it started outside an hour later with a cookout and toasts to the birthday girl.

"They were nice people drinking a glass of wine," said Roxie Means,?who attended the party Friday her 14-year-old daughter, Cassie. "They weren't drunk. They weren't overdoing anything."

And before she left, Carter-Camp's two children wanted to know if she would be back on Saturday.

"I was telling the kids good night," Cassie Means said. One of the children asked her, "'Cassie, are you coming over to play with us tomorrow?' I said, 'yeah.'"

The child continued, "'you promise me you'll be here tomorrow?'" Cassie Means recalled. "I said, 'I promise you I'll be here when you wake up to play with you. I'll be here right when you wake up."

Hours later, the only adult survivor was smoking a cigarette outside, noticed the fire and came running to Means' home and started "beating down the door," Roxie Means said.

Investigators believe that all of the victims were asleep at the time of the fire and died of smoke inhalation.

Jones said?it?was the worst house fire Charleston has ever seen in terms of loss of life. In 1949,?a?fire at the Woolworth department store in downtown Charleston killed seven firefighters, according to the Gazette.

James Cameron makes final preparations for historic deep-sea dive

Weather permitting Saturday, explorer and filmmaker James Cameron could take his Deepsea Challenger to the bottom of the world, a place of perpetual cold, darkness, and abiding mystery.?

There was none of the usual Hollywood fanfare as James Cameron pulled up anchor and headed out to sea with his cast of adventurers last Monday on the final, spectacular leg of what will arguably be the greatest production he has ever worked on.

The departure of the?Mermaid Sapphire and Barakuda expedition vessels from Apra Harbor Guam, set the famously thrill-seeking film director on course to dive in a 12-ton submersible to the bottom of the world, a place of perpetual cold and darkness, life-threatening depth, and abiding mystery.

Inclement weather has kept Cameron from his goal over the last few days, prolonging the suspense and forcing him and his team of more than 60 support workers, technical advisers, scientists and family to wait it out for most of the week on the tiny Pacific atoll of Ulithi.

But with a possible window in the weather Saturday, the big moment could now be just hours away. Cameron, 57, and his?Deepsea Challenger?are ready to roll, he announced through National Geographic, his expedition partners, early this morning.

The journey will, if all goes well, take him seven miles below the surface of the Pacific Ocean to Challenger Deep, the lowest known point on Earth, located in the Mariana Trench south-west of Guam.

If he descends safely, he will be the first person to complete the journey alone to what he describes as ?a place less understood, more forbidding and perhaps less forgiving than the farthest reaches of space.?

Among his advisers currently aboard the?Mermaid Sapphire?mother-ship is a man who might be considered the ultimate authority on Challenger Deep, retired US Navy Captain Don Walsh, 80, who took the plunge aboard the steel bathyscaphe?Trieste?in 1960 with Swiss oceanographer Jacques Piccard.

No one had made the 36,070-foot trip before and no one has done it in the 52 years since.

What can Cameron expect? ?For us, it was rather like a foggy day in your car, where you turn the lights on and the fog comes back in your face,? says Capt. Walsh, whose?Trieste?crewmate died four years ago.

?As you descend, you lose your natural light fast and you?re in the abyss pretty quick ? no sunlight penetration at all. But if you turn off the lights, there?s this host of bio-luminescence generated by creatures of the abyss. It?s not that bright, like little snowflakes with light reflecting off them. As you pass down through them, the illusion is that they?re streaming up from below.?

Despite measuring 24feet in total,?Deepsea Challenger?affords its 6 foot, 2 inch pilot a space just 43 inches in diameter; the bigger one builds the pressurized compartment, the thicker its walls need to be and, therefore, the heavier the sub will become. Smaller equals lighter, equals slicker.

The water pressure will be so great ? eight tons per square inch ? that the sub?s dimensions will shrink by around 2.5 inches (6.3 centimeters) on the way down.

Cameron ?has been practicing yoga to help him fold his frame into the cramped, spherical space and maintain his position for nine hours ? 90 minutes to two hours for the descent, six hours exploring the sea floor, and an hour back up to the surface.

Gene expression abnormalities in autism identified

ScienceDaily (Mar. 23, 2012) ? Genetic studies find dysregulation in pathways that govern development of the prefrontal cortex in young patients with autism

A study led by Eric Courchesne, PhD, director of the Autism Center of Excellence at the University of California, San Diego School of Medicine has, for the first time, identified in young autism patients genetic mechanisms involved in abnormal early brain development and overgrowth that occurs in the disorder. The findings suggest novel genetic and molecular targets that could lead to discoveries of new prevention strategies and treatment for the disorder.

The study to be published on March 22 in PLoS Genetics uncovered differences in gene expression between brain tissue from young (2 to14 years old) and adult individuals with autism syndrome disorder, providing important clues why brain growth and development is abnormal in this disorder.

Courchesne first identified the link between early brain overgrowth and autism in a landmark study published by the Journal of the American Medical Association (JAMA) in 2003. Next, he tested the possibility that brain overgrowth might result from an abnormal excess of brain cells. In November 2011, his study, also published in JAMA, discovered a 67 percent excess of brain cells in a major region of the brain, the prefrontal cortex -- a part of the brain associated with social, communication and cognitive development.

"Our next step was to see whether there might be abnormalities of genetic functioning in that same region that might give us insight into why there are too many cells and why that specific region does not develop normally in autism," said Courchesne.

In the new study, the researchers looked towards genes for answers, and showed that genetic mechanisms that normally regulate the number of cortical neurons are abnormal. "The genes that control the number of brain cells did not have the normal functional expression, and the level of gene expression that governs the pattern of neural organization across the prefrontal cortex is turned down. There are abnormal numbers and patterns of brain cells, and subsequently the pattern is disturbed," Courchesne said. "This probably leads to too many brain cells in some locations, such as prefrontal cortex, but perhaps too few in other regions of cortex as well."

In addition, the scientists discovered a turning down of the genetic mechanisms responsible for detecting DNA defects and correcting or removing affected cells during periods of rapid prenatal development.

Autism is a highly heritable neurodevelopmental disorder, yet the genetic underpinnings in the brain at young ages have remained largely unknown. Until now, few studies have been able to investigate whole-genome gene expression and genotype variation in the brains of young patients with autism, especially in regions such as the prefrontal cortex that display the greatest growth abnormality.

Scientists -- including co-first authors Maggie Chow, PhD, and Tiziano Pramparo, PhD, at UC San Diego -- identified abnormal brain gene expression patterns using whole-genome analysis of mRNA levels and copy number variations from 33 autistic and control postmortem brain samples. They found evidence of dysregulation in the pathways that govern cell number, cortical patterning and cell differentiation in the young autistic prefrontal cortex. In contrast, in adult patients with autism, the study found that this area of the brain shows dysregulation of signaling and repair pathways.

"Our results indicate that gene expression abnormalities change across the lifespan in autism, and that dysregulated processes in the developing brain of autistic patients differ from those detected at adult ages," said Courchesne. "The dysregulated genetic pathways we found at young ages in autism may underlie the excess of neurons -- and early brain overgrowth -- associated with this disorder."

Dog survives 53 days in wild, reunited with owner

RENO, Nev. (AP) ? Barbara Bagley says she never gave up hope that her dog would be found alive in the Nevada desert after the animal bolted from the scene of a crash that critically injured her and killed her husband.

But the Salt Lake City woman endured plenty of frustration until her beloved 4-year-old Shetland sheepdog, Dooley, was tracked down Feb. 18 after surviving 53 days in the wild on roadkill and scattered ranch water sources.

"I would think about Dooley constantly," she said. "There were TV commercials with dogs that made me think about him and cry. He's just the sweetest dog."

The Dec. 27 single-vehicle accident on Interstate 80 near Battle Mountain, about 225 miles east of Reno, sent Bagley and her 55-year-old husband, Brad Vom Baur, to the hospital in critical condition. Their other sheltie, Delaney, was killed in the wreck. Dooley ran away and vanished.

Bagley, 48, suffered a concussion, broken ribs, a shattered wrist and two punctured lungs. As soon as she mustered up enough strength, she turned her attention to a search for her dog in the sprawling sage-covered plains and hills of northeastern Nevada.

Realizing what Dooley could mean for her recovery, dozens of Nevada volunteers responded to a Facebook plea for help in looking for him. But the search was canceled before it began after the Jan. 6 discovery of what appeared to be the dog's remains along the interstate. The same day, her husband died.

"It was a horrible day for me," Bagley recalled. "But something inside me told me Dooley was still alive out there. I wasn't 100 percent sure, but I didn't grieve for Dooley like I did for my husband and our other dog."

More than three weeks later, Bagley's spirits were buoyed after a woman reported spotting "a Lassie-type" dog near the accident scene. A subsequent search joined by Bagley turned up nothing, but a railroad crew spotted a dog matching the same description in mid-February in the same area about 15 miles east of Battle Mountain.

Further searches netted a positive identification of Dooley but frustration as well because the skittish dog kept fleeing from Bagley and other searchers. Finally, Shannon Sustacha of Lamoille, who was on horseback, and a Bagley friend driving a Jeep cornered Dooley only five miles from the crash scene. The friend managed to nab the sheltie and put him in the Jeep.

An ecstatic, tearful Bagley arrived at the scene a short time later.

"Barbara got next to us and said three times, 'You think he'll remember me?'" Sustacha said. "When Barbara opened the door and looked at him, she said, 'My beautiful boy, my beautiful boy, you're home.' Oh, boy, all of us cried. I knew his adventure in Nevada was over. I also knew he and Barbara could start healing together."

A short time later, an exhausted Dooley sat on his owner's lap in the Jeep and fell asleep. He later began following Bagley around.

"I was overjoyed that I was going to have him back in my life. I think he felt the same about me," she said.

During his ordeal, Dooley's weight dropped from 44 pounds to 20 pounds. He was once spotted devouring a dead coyote along the roadway. A long bird bone was pulled from his throat by a veterinarian.

Since then, the dog has gradually put on weight and resumed regular walks with his owner. While Bagley is still going through the grieving process over her husband's death and recovering from her injuries, Dooley's presence has picked up her spirits immensely.

"He's the physical and mental affection that I need to recover," she said. "I owe him so much for the hope I have now and the renewed faith I have in prayer. Dogs are so great because of their unconditional love."

Bagley, a phlebotomy supervisor at LDS Hospital in Salt Lake City, also thinks her husband had something to do with Dooley's safe return home.

"It's a message from my husband who was looking out for him," Bagley said. "It was a miracle that we got Dooley. He couldn't have survived much longer out there."
